Northumbrian Water said there was no flooding in the streets or in people's homes around the sewer, near the junction of Newlands Avenue, Stockton-on-Tees.
The A139 Norton Road was closed at Norton Avenue on Monday evening.
"We believe that this will be a complex repair, and our teams will be working around the clock for the next few days to allow the road to be reopened as soon as possible," a spokesperson said.
Northumbrian Water said it was still investigating the cause.
BBC/ Louise HobsonDiversions were put in place around the busy road.
"Due to the location of the collapsed sewer, which is near the junction of Newlands Avenue, we have put a road closure in place on Norton Road in order to allow repairs to be carried out safely and to keep pedestrians and road users safe," Northumbrian Water said.
